Abstract BRS Rendeira and BRS Progresso are cassava (Manihot esculenta Crantz) cultivars with tolerance to post-harvest deterioration and high mean yields of fresh root (31.1 and 27.9 t ha-1) and starch (8.9 and 8.1 t ha-1) in annual production cycles. Both new varieties are recommended for the flour and starch industry.

Abstract The objective of this work was to evaluate the genetic diversity of Xanthomonas phaseoli p v. manihotis (Xpm) from eight populations from five cassava producing states in Brazil, through the rep-PCR (BOX-PCR and ERIC-PCR) and variable number of tandem repeat (VNTR) markers. Cassava leaves with symptoms of cassava bacterial blight were collected in eight municipalities, and the Xpm isolates were identified by amplification with primers specific for these isolates. The identity of the Xpm isolates was confirmed with the BOX-PCR, ERIC-PCR, and VNTR markers. The observed selection pressure, together with the mode of reproduction and the mechanisms that increase genetic variability, allows of the pathogen populations to adapt according to microclimate variation, contributing to a differentiated reproductive success. ERIC-PCR and VNTRs are the best markers for evaluating the genetic variability in the eight studied Xpm populations. However, ERIC-PCR is the marker that best separated the groups by population and presented a higher similarity between the isolates of the same population. The study of the genetic diversity of Xpm is key to improve disease monitoring and management strategies in cassava crops.

Resumo O objetivo deste trabalho foi avaliar a diversidade genética de Xa nthomona s phaseoli p v . ma nihot i s (Xpm) de oito populações de cinco estados produtores de mandioca no Brasil, por meio de marcadores rep-PCR (BOX-PCR e ERIC-PCR) e variable number of tandem repeats (VNTRs). Folhas de mandioca com sintomas de crestamento bacteriano foram coletadas em oito municípios, e os isolados Xpm foram identificados por amplificação com iniciadores específicos para esses isolados. A identidade dos isolados Xmp foi confirmada com os marcadores BOX-PCR, ERIC-PCR e VNTRs. A pressão de seleção observada, junto com o modo de reprodução e os mecanismos que aumentam a variabilidade genética, permite que as populações do patógeno se adaptem de acordo com a variação dos microclimas, o que contribui para o sucesso reprodutivo diferenciado. ERIC-PCR e VNTRs são os melhores marcadores para avaliar a variabilidade genética das oito populações Xpm estudadas. No entanto, ERIC-PCR é o marcador que melhor separou os grupos por população e apresentou maior similaridade entre os isolados de uma mesma população. O estudo da diversidade genética de Xpm é fundamental para delinear estratégias de manejo e monitoramento de doenças na cultura da mandioca.

Abstract The new cassava variety ‘BRS Novo Horizonte’ was developed and released in Bahia, Brazil for industrial use, particularly for starch production. The fresh and dry root yields were 39% and 44% higher, respectively, than those of local varieties. The root quality traits allowed the extraction of high-quality starch. The variety is also adapted for mechanized cropping systems.

ABSTRACT. Modifications were made in the PCR conditions aiming to overcome the problem of non-amplification of the Xanthomonas phaseoli pv. manihotis (Xpm) fragment, using the primer pair XV / XK described in the literature. The objective of this study was to propose changes in the primers already described (XV / XK_MOD) and validate the use of these new primers in identifying Xpm. The validation procedure was carried out with the primer pair XV and XK_MOD, using different strains of Xpm, other plant pathogenic and endophytic bacteria genera and one isolate of X. phaseoli pv. passiflorae. As a control, additional reactions were conducted in multiplex with the universal primers for the 16S rRNA gene of the bacteria together with XV / XK and XV / XK_MOD. Using the forward primer (XV) described in the literature together with the modified reverse primer (XK_MOD), it was possible to achieve amplification from DNA extracted from in vitro cultures and from infected tissue, but no amplification was noticed for the primer pair described in the literature, confirming the effectiveness of the proposed modification.

ABSTRACT Cassava root rot diseases such as dry and black root rot are listed among the major threats, since its affect the main product (tuberous roots), causing a progressive decline in yield and affecting subsequent crop cycles, being the use of resistant varieties the most reliable control measure on field level. The objective of this study was to identify inoculation methods for the early evaluation of genotypes, considering the level of resistance to dry (DRR) and black (BRR) root rot diseases. Different methodologies and plant tissues were evaluated, based on the immersion of cassava tissues (roots and stem cuttings), soil infestation, and inoculation of detached tissues (leaves, tuberous roots and stem cuttings). The following parameters were evaluated for inoculations based on tissue immersion: disease index (ω); aerial part weight (g); fresh weight of the roots (g); and volume (cm3). For the inoculations on detached tissues, the percentage of lesioned area was determined. Immersion methods for roots and lesioned stems did not show typical symptoms of DRR and BRR during the two-month evaluation period. The soil infestation method did not differ from the stem immersion method as to the reduction of aerial part weight and the disease index, whereas both can be recommended for resistance selection trials. There was a positive correlation between the BRR and DRR data for the stem inoculation (r = 0.94, p = 0.001) and for DRR in the peel and root pulp (r = 0.73, p = 0.05). Therefore, the resistance within each tissue is apparently independent and should be compared with the behavior of the genotypes in the field.

RESUMO O murcha de Fusarium, causado por Fusarium oxysporum f. sp. cubense (FOC), é uma das principais doenças da cultura da bananeira. A principal estratégia de controle da doença faz-se com o uso de variedades resistentes, sendo que, para a sua obtenção, faz-se necessário identificar genótipos para utilização nos programas de melhoramento genético da cultura. O objetivo do presente estudo foi identificar fontes de resistência em acessos do Banco Ativo de Germoplasma (BAG) de Bananeira da Embrapa Mandioca e Fruticultura. Foram avaliados 41 acessos do BAG, sendo 17 diploides, 21 triploides e dois tetraploides. Também foram avaliados a área abaixo da curva do progresso da doença, o índice interno da doença e o período de incubação. Há variabilidade genética disponível entre os acessos do Banco de Germoplasma de Bananeira para resistência ao FOC, com destaque à imunidade expressada pelo genótipo M53. Os acessos Birmanie, PA Songkla, Pirua, Imperial, Poyo, Ambei, Walebo e Kongo FRF 1286 expressaram resistência.

ABSTRACT Fusarium wilt (syn= Panama disease), caused by Fusarium oxysporum f. sp. cubense (FOC), is one of the most destructive diseases of banana, being genetic resistance the main management strategy for this disease. Since the pathogen is constantly evolving to supplant the genetic resistance, new sources of resistance must be investigated by genetic improvement programs aiming to developing new varieties. The objective of the present study was to identify sources of resistance from the different accessions maintained in the banana active germplasm bank (BAGB) at Embrapa Mandioca e Fruticultura. Forty-one BAGB accessions were evaluated, including 17 diploids, 21 triploids, and two tetraploids. The area under the disease progress curve, disease index, and incubation period were also evaluated. In relation to FOC resistance, there is genetic variability available among the BAGB accessions. The genotype M53 is notable for the complete resistance it expressed, and the accessions Birmanie, PA Songkla, Pirua, Imperial, Poyo, Ambei, Walebo, and Kongo FRF 1286 expressed quantitative resistance.

ABSTRACT: Although root rot is one of the major diseases affecting Brazilian cassava (Manihot esculenta Crantz.), little is known about the diversity of root rot pathogens. In this study, diseased plants exhibiting root rot symptoms were collected from cassava-producing regions in five Brazilian states: Bahia, Sergipe, Paraíba, Maranhão, Tocantins, and Paraná. Seventy isolates were obtained and assigned to species complexes based on rDNA's ITS (internal transcribed spacer of the ribosomal DNA) region (ITS1, ITS2 and 5.8S). A total of seven species complexes was found belonging to the genus Fusarium (56/74), followed by Lasiodiplodia (8/74), Neoscytalidium (6/74), and Diaporthe/Phomopsis complex (2/74), Phytophthora, and Corallomycetella (1/74 each). These species were distributed differently according to sample locations and states, but overall, the F. solani species complex (FSSC) was the most prevalent. A number of phylogenetic lineages had not been previously reported as being associated with cassava-root rot disease, such as: F. graminearum (FGSC), F. incarnatum-equiseti (FIESC) and F. chlamydosporum (FCSC) complexes, and a phylogenetic lineage most closely related to P. phaseolorum. Results suggest the need to improve knowledge of the species associated with cassava, including multilocus phylogeny for a more specific characterization, and differences in the resistance background associated with these species, as a strategy to incorporate resistance to multiple pathogens in cassava breeding programs.

RESUMO O conhecimento da variação fenotípica do germoplasma de mandioca (Manihot esculenta Crantz) permite estimar a variabilidade genética como suporte para seleção de genitores contrastantes. O objetivo deste trabalho foi definir grupos homogêneos de germoplasma de mandioca com base em características produtivas, resistência a doenças e qualidade de raízes utilizando o método não-hierárquico K-means. Os valores genotípicos estimados pelo BLUP (Best Linear Unbiased Predictor) foram utilizados para a análise de agrupamento. O número de grupos foi definido no início da estabilização da menor soma de quadrados dentro dos grupos. Foram obtidos 17 grupos para representar a diversidade do germoplasma, cujo número de acessos variou de 7 (Grupo 15) a 69 (Grupo 9). Em geral, os acessos pertencentes aos grupos 1; 4; 7; 12; 15 e 16, apresentaram boas características agronômicas, tais como alta produtividade de raízes frescas e de amido (> 60,7 t ha-1 e 18,6 t ha-1, respectivamente). Por outro lado, apenas o grupo 15 apresentou menor severidade de bacteriose. Os grupos obtidos apresentaram grandes diferenças entre si, conforme valores da soma de quadrados dentro dos grupos, que variavam entre 215,1 (Grupo 15) a 2338,3 (Grupo 8). O K-means permitiu a formação de grupos consistentes com base nas características agronômicas avaliadas. Portanto, o algoritmo K-means foi eficiente para a formação de grupos homogêneos com baixa variação genotípica dentro, especialmente para análises de grandes quantidades de dados, como em bancos de germoplasma de mandioca.

ABSTRACT The knowledge of the phenotypic variation of cassava (Manihot esculenta Crantz) germplasm allows the estimative of the genetic variability to support the selection of contrasting genitors. Therefore, the aim of this work was to define homogeneous groups of cassava germplasm based on yield traits, disease resistance and root quality using K-means as a non-hierarchical method. Breeding values estimated by Best Linear Unbiased Predictor (BLUP) were used for the cluster analysis. The number of groups was defined according to the stabilization of the smallest within-group sum of squares. Seventeen clusters were defined to represent the diversity of the germplasm, whose number of accessions ranged from 7 (Group 15) to 69 (Group 9). In general, accessions belonging to Groups 1, 4, 7, 12, 15 and 16 showed good agronomic traits, such as high fresh root yield and starch yield (> 60.7 t ha-1 and 18.6 t ha-1, respectively). In contrast, only Group 15 presented low bacterial blight severity. The groups obtained showed strong differences, as evidenced by the within-groups sums of squares values, which ranged from 215.1 (Group 15) to 2,338.3 (Group 8). The K-means algorithm allowed the formation of consistent groups based on yield traits, disease resistance and root quality. Therefore, the K-means algorithm was efficient in the formation of groups with low within genotypic variation, especially concerning large amounts of data, such as in cassava germplasm banks.

O objetivo deste trabalho foi avaliar, por meio de marcadores SSR, a diversidade genética de Xylella fastidiosa no Estado da Bahia. Foram estudadas duas das principais regiões produtoras de citros no Estado, o Litoral Norte e o Recôncavo Sul. Para fins comparativos, utilizaram-se dez amostras provenientes do Estado de São Paulo. Foram empregados os seguintes iniciadores: ASSR20, OSSR9, OSSR17, CSSR4, CSSR12 e CSSR20, dos quais os quatro últimos permitiram identificar 22 loci polimórficos. As populações de X. fastidiosa presentes em citros no Estado da Bahia apresentam elevada diversidade genética, com base nos marcadores SSR, com pools gênicos distintos e agrupamento geográfico. No Litoral Norte, as populações do isolado apresentam maior diversidade genética do que as da região do Recôncavo Sul da Bahia.

The objective of this work was to evaluate, by SSR markers, the genetic diversity of Xylella fastidiosain the state of Bahia, Brazil. Two of the main citrus producing regions of the state were evaluated, Litoral Norte and Recôncavo Sul. Ten samples from the state of São Paulo, Brazil, were used for comparison. The following primers were used: ASSR20, OSSR9, OSSR17, CSSR4, CSSR12, and CSSR20, of which the last four allowed the identification of 22 polymorphic loci. The citrus populations of X. fastidiosa in the state of Bahia have high genetic diversity, based on SSR markers, with distinct gene pools and geographical grouping. In Litoral Norte, the populations of the isolate show higher genetic diversity than those in the Recôncavo Sul region of Bahia.

O objetivo deste trabalho foi identificar fontes de resistência à podridão seca das raízes ocasionada por Fusarium sp. em acessos de mandioca. Inoculou-se uma suspensão (20 µL) de macroconídios de 11 isolados de Fusarium sp. em raízes de 353 acessos e sete variedades comerciais de mandioca. Dez dias após a inoculação, avaliou-se a área total colonizada pelo patógeno na polpa das raízes, por meio de análise de imagens digitais. A análise de agrupamento revelou a presença de cinco grupos quanto à resistência. As áreas lesionadas das raízes variaram de 18,28 a 1.096,07 mm² nos acessos BGM 1518 e BGM 556, respectivamente. Os genótipos BGM 1042, BGM 1552, BGM 1586, BGM 1598 e BGM 1692 apresentam as melhores características agronômicas.

The objective of this work was to identify sources of resistance to dry root rot induced by Fusarium sp. in cassava accessions. A macroconidial suspension (20 µL) of 11 Fusarium sp. isolates was inoculated in cassava roots, from 353 acessions plus seven commercial varieties. Ten days after inoculation, the total area colonized by the pathogen on the root pulp was evaluated by digital image analysis. Cluster analysis revealed the presence of five groups regarding resistance. The root lesion areas ranged from 18.28 to 1,096.07 mm² for the accessions BGM 1518 and BGM 556, respectively. The genotypes BGM 1042, BGM 1552, BGM 1586, BGM 1598, and BGM 1692 present the best agronomical traits.

This study aimed to characterize the resistance of 31 passion fruit (Passiflora sp.) genotypes to Fusarium oxysporum f. sp. passiflorae (FOP). About 20 seedlings of each genotype were inoculated by immersing the roots in a suspension of 10(6) macroconidia mL-1 for five minutes and then transplanting them into pots containing a mixture of soil and sterile substrate. They were evaluated daily for 120 days for the occurrence of wilt symptoms and death. These data were used to calculate the area under the disease progress curve, for survival analysis and grouping. The mortality rate ranged from 0 to 100% and the AUDPC ranged from 0 for genotypes without symptoms of the disease to 6,650.63 for the hybrid HFOP-01. Genotypes were classified into four subgroups: resistant, moderately resistant, susceptible and highly susceptible. The resistance was genotype dependent, but no significant variation was found between the purple and the yellow P. edulis.

Fusarium oxysporum f. sp. passiflorae W.L. Gordon (FOP) is one of the most important fungal pathogens of passion fruits. Understanding molecular variation of isolates from different areas is of utmost importance. Molecular fingerprinting on 14 isolates of FOP were conducted using AFLP molecular markers (Amplified Fragment Length Polymorphism), and their genetic variability were estimated. Twenty-five AFLP primer combinations were selected for amplification of FOP isolates and one for Fusarium oxysporum f. sp. cubense W.C. Snyder & H.N. Hansen (FOC), resulting in 99% polymorphic fragments, with an average of 40 fragments per primer combination. Specific fingerprints could be generated for most of the isolates evaluated; we observed a high power of discrimination of the AFLP primer combinations, with the presence/absence of up to 26 specific fragments per isolate. Thus, specific fingerprinting was obtained for 10 of the 15 isolates analyzed. The values of the polymorphic information content, the index and the resolving power of the markers showed wide variation and reflected the high informative contents of the primers used in the characterization of the FOP isolates. The FOP isolates were divided into four groups, irrespective of their geographic origins, with the allocation of 5, 7, 1 and 1 FOP isolates into Groups II, III, IV and V, respectively. A wide genetic diversity was observed in FOP isolates, which should be taken into consideration when implementing strategies for the improvement of passion fruit in the search for cultivars with multiple resistance to different isolates.
